Na... maybe they are already working just fine all the time. Try again plug in an usb device and make sure to plug in the usb-connector the right way and not upside down. Your mouse probably needs a PC-reboot before it works.

You seem to have missed what I am trying to say.
The front USB ports do not work, period.

I have tried the new mouse, a USB memory stick, and a USB external 3.5in diskette drive.

Front USB ports:
None of them work in either of the front USB ports, windows detects nothing, the mouse LED does not even come on.

Rear USB ports:
All of them are detected with windows making a tone the moment they are plugged into a rear USB port, and they work with no trouble.

That's the whole problem.
They appear to be enabled in BIOS
They appear to be plugged firmly into the MB.
The USB stuff that is listed in the Device Manager shows now problems.
Despite all of that, the front USB simply do not work.

OMFG...

Some time with Google and my MB manual reveals the computer shop goofed big time.
I decided to trouble shoot by just removing the cable and putting it into a different USB slot on the MB.
Then I noticed something when removing my sound card to get a clear space to get at the USB cable.
It was plugged in alright...
It just was not plugged in right...

This is where I tried to point out how the plug was in the slot. I should have added this info, it would have saved us both some trouble

The second should have been more like this.

One thing I had tried to note was that there were 2 pins to the left of the plug.
Basically... The USB cable was off to the right by a whole pin!

After removing the sound card, I was able to get it loose, and to be on the safe side, when I put it back in, I put it in one of the other USB slots on the MB.

After reboot, a quick check with the USB external diskette drive quickly led to a tone and Windows recognizing the device.

The only complication during the fix was getting ready to get the PC hooked back up, going to plug in the speakers and noticing my sound card was on the desk, so I had to reopen the case to put it back in...

I find it odd that the USB cable was off by a whole pin.
I also find it odd that I did not know any better than to notice that exposed pins was not right.
I also find it more odd the USB port on the MB is wide enough for the USB cable to end up being off by a whole pin.

Now if I have to get out my backup USB keyboard, I will still have free (working) USB slots for other things like my USB memory sticks.

Now I can goof off with, er, play test more with my Sims 3 graphics settings for the other topic.

That's your case manufacturer to blame. My USB "blocks" from the Coolermaster Stacker have the same shape and wideness as the receiving connectors on the motherboard, filling the hole exactly; one pin off wouldn't be possible.
That reminds me: all Asus boards have a few USB extension blocks, yours should have too. They're blue. You fit the too small case-"block" into the blue Asus block, which then fits exactly into the receptacle on the motherboard.
